Introduction
Hybridoma sequencing is generally based on reverse transcription polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends (RACE) technologies, which rapidly amplifies the 5’ and 3’ ends of cDNA from a low number of transcripts, and is widely used because it is simple, fast and inexpensive.First, we will extract your mRNA from your hybridoma cells and conduct RT-PCR to obtain the cDNA. Then, the light chain, heavy chain, or full-length fragment will be amplified by PCR or 3’ and 5’ RACE. The fragments can be connected to T-vectors and sequenced.
